The parental T47D cell line was established from the pleural effusion of a ductal carcinoma of the breast. The tumour cell line, T47D-E2J, was created as a breast cancer model with an O-linked glycosylation profile similar to normal mammary epithelial cells. T47D-E2J cells overexpress the enzyme C2GnT1, a beta-1,3galactosyl-O-glycosyl-glycoprotein. C2GnT1 is the dominant enzyme expressed in normal breast tissue known to convert core 1 glycoproteins to core 2 glycoproteins, by the addition of N-acetylglucosamine to N-acetylgalactosamine. Expression of the C2GnT1 enzyme has been found to be decreased or absent in most breast cancer cell lines despite still expressing mRNAs coding for this enzyme. Together with wild-type T47D and T47D-STn these cell lines represent a panel of isogenic cells with different O-linked glycosylation patterns. |
