Relevance | NOD1 responds to Helicobacter pylori infection in a Type IV secretion system-dependent manner. H. pylori causes chronic inflammation, a key precursor to gastric adenocarcinoma. AGS cells deficient in NOD1 (as generated by CRISPR) can be used to model gastric adenocarcinoma in the context of various pathogens. NOD1 regulates host epithelial cell responses. Upregulates antimicrobial peptides of the beta-defensin (hBD) family secreted by epithelial cells. |
Production Details | CRISPR/Cas9 gene editing was used to generate NOD1 knock out clones. AGS cells were transfected with vectors expressing “Cas9 nickase” nuclease and pairs of guide RNAs (gRNAs) specific for the NOD1 gene. Clone 41A8 was confirmed as having a 71 base pair deletion within the coding regions of the caspase activation and recruitment domain (CARD) of NOD1. |
