Relevance | MPO is a mediator enzyme secreted by inflammatory cells e.g. activated neutrophils and monocytes. MPO produces hypochlorous acid (HOCl) from hydrogen peroxide (H2O2) and chloride anion (Cl-) (or the equivalent from a non-chlorine halide) during the neutrophil's respiratory burst, requiring heme as a cofactor. Furthermore, it oxidizes tyrosine to tyrosyl radical using hydrogen peroxide as an oxidizing agent (Heinecke et al., 1993). Both hypochlorous acid and tyrosyl radical are cytotoxic and are used by neutrophils to kill bacteria and other pathogens. High levels of MPO have been reported in atherosclerotic plaques (Blake and Ridker, 2003) and accumulating evidence indicates that MPO may also have a causative role in plaque vulnerability (Hazen, 2004). MPO can generate several reactive oxidised intermediates, concomitantly inducing oxidative damage of cells and tissues (Brennan et al., 2003). |
